HUwaaaa~ busynye mggu ni~ maklumla sbb last week kan... next week dah study week.. then final week!!! owh~ tira ta sedie lagi!!!! huwarggghh~ mcm mne ni! mcm mne nih!!! hehe~ tira cool~ cool~ cool~ sejuk~ heee~ ok2 ... dah okeh dah... hehe... hari ni letih ckt coz td buat lab crossmatch~ ala... crossmatch~ tak tahu?? tak tau ye?? hehe~ padan2 kan darah~ hah~ skunk mesti dah tau kan! kan! heee... oklah na cite cikit... actually... crossmatch ni penting untuk blood tranfusion... sebelum kt bg darah or derma darah kat org tu... kt kene chek dulu, sama ade darah tu COMPATIBLE atau INCOMPATIBLE... walaupown darah jenis O nak derma kat O sekalipun... kt kene chek COMPATIBALITY dye tau! tak boleh men tranfuse2 je~ nnt kang noktah pulak nyawa org tu~ huhu... oklah kat sini tira nak cite sikit ttg ABO blood group yg mane boleh derma.. dan yang mane boleh terime... untuk pengetahuan anda semua [ceh ,buat ayat POYO la pulak... hehe ^^]... oklah kalau anda darah O, anda boleh derma kepada semua jenis darah tau! A,B,AB dan juge O!! kirenye,org darah O ni mmg sgt "PEMURAH"wah~ bangge2!! sbb tira darah O!! tp malangnye, bile kumpulan O ni kurang darah, kt hanya boleh terime darah O shj~ kesian kan! kan!hehe... oklah kalau utk darah2 yang laen ni... sile rujuk kat table ni ye~

oklah sekarang berbalik kepada crossmatch... huh~ mmg kene penuh kesabaran nak buat crossmatch ni... ttd pown slow je nak buat... huhu... oklah kat sini tira na share procedure mcm mane nak buat crossmatch.Procedure ni iyelah standard procedure yg tira gunekan utk buat lab test td... sorrylah,dlam bahase inggeris lah ye... almaklumlah kan... tira org UK[ulu kinta je....^^]
Procedures:
A. Protein Phase( Saline Phase)
1. Wash donor’s cell 3 times with saline.
2. Prepare a 5% suspension of donor’s cell in saline.
3. Add 2 drops of recipient’s serum to each of two tubes labelled i&ii
4. Add 1 drop of 5% donor cell suspension to each tube
5. Add 2 drops of Ionic strength solution(LISS)/ OR ALTERNATIVELY CAN USE 22% BOVINE ALBUMIN) to tube (i)
* BOVINE ALBUMIN – incubate for minimum 30 minutes and LISS 10 minutes.
6. Centrifuge both tubes for 1 minute at 1000 rpm.
7. Examine both tubes macroscopically for agglutination and/or haemolysis.
8. If both tubes are free from agglutination/ haemolysis proceed to thermo phase.
*Washing- 3400rpm, 1 minute
*Reading – 1000rpm,1 minute or 3400rpm,15 seconds
B. Albumin Phase( Thermo phase)
1. Incubate both tubes at 37°C for 10 mins or 30 mins minimum if 22% BOVINE ALBUMIN is used.
2. Centrifuge both tubes for 1 min @ 1000rpm.
3. Examine both tubes macroscopically and microscopically for agglutination or haemolysis.
4. If negative for both, proceed to Anti Human Phase.
C. Anti –Human Phase
1. Carefully dislodge all cells from the bottom of tubes and fill with saline.
2. Centrifuge at 3400 rpm for 1 minute and decant.
3. Decant the saline as completely as possible after the last work.
4. Add 2 drops of AHG and mix well.
5. Centrifuge for 1 minute at 1000rpm.
6. Examine for agglutination macroscopically and if negative and if negative under low power microscope.
7. If no agglutination is present, add 1 drop of COOMB’S CONTROL CELLS (CCC) to confirm the crossmatch as VALID.
8. If CCC shows positive result, the test is satisfactory and the blood can be issued as COMPATIBLE.
hah... rumitkan! rumitkan! huhu... mmg la rumit, sbb crossmatch ni penting tau!! kt kene buat secara berhati-hati.. kalau tersilap interprate result...dan kt maen tranfuse je darah tu kat org laen, dan bile body org tu reject, boleh membawa maut!! HABISLAH!! HABISLAH~jd kt kene buat crossmatch dgn betul2!!! jgn maen2... huhu... kat sini, mesti anda tertanye2
1.brape lame nak tggu buat crossmatch ni?? bile mase na tranfuse darah kat org laen??
2. mcm mane kalau kes accident, takan nak tggu berjam2 utk serasikan darah ni baru boleh tranfuse darah kat org tuh!??

1.hmm... crossmatch ni dibuat dlm mase lbh kurg 45 minit.. berdasarkan sumber internet lah... dan kt bukan sahaja crossmatch sorg donor je... selalunye kt akan crossmatch dengan 3 atau 4 sekali donor... sbb nak tgk, donor mane yg darah dye "serasi bersama" dengan recipient tuh...sbb kalau buat 1 donor je, nnt donor tu tak MATCH dgn recipient tu, kan kene buat lg crossmacth yg laen... ambil mase kan...jd itu sbbnye kt kene buat dgn byk donor.
2.ok time accident!! mmg emergency kes lah kan.. takan la na tggu sampai 45 minit buat crossmatch ni... nnt belum sempat cross2 , dah melayang nyawa patient tuh... ok selalunye, bile time accident... kt akan gunekan darah O .. sbb kan O universal donor, kt terus tranfuse tanpa buat crossmatch... ini utk emergency kes lah!! serasi tak serasi darah tu kt tgk kemudian... lepas tranfuse,then lepas tu bru lah kt selesaikan masalah serasi tak serasi ni...
oklah~ wah kali ni post tira terlebih panjang la pulak!! aigoo~ hehe...dan inilah dye... care2 nak buat crossmatch!! test yang paling penting dalam immunohematology lab~ huhu ^^

heee... oklah, sbnrnye sng je na buat mac conkey agar ni.. cume tu lah, bau dye~ YA ALLAH~ memang menyelerakan~ huuuu... dan disebabkan tu lah tira jd allergic! huhu.... ok2...care na buat dye sng je.... kt, set up sume peralatan... kemudian... timbang mac conkey agar, gune balances td... lps tu... masukkan ke dlm beaker dan campurkan dgn distilled water... kemudian panaskan dye hingga menggelegak!! huhuuuu.... nnt kan colour dye akan jd spti warna air ribena yg sgt2 menyelerakan~ huuu... [tp bau!! MASYAALLAH~~~ ]
dan setelah iyenye mendidih sepenuhnye... alihkan api.. dan sejukkan sedikit agar td tau... huhu... lps tu, masukkan dlm autoclave bottle dan autoclave selama 20 minit... [kalau ta clap... heee~ ;pp].. lps autoclave... kt tuangkan agar td ke dlam petri dish!sejukkan dan dah siap! heee~ siaplah AGAR2 MAC CONKEY KITA!!!agar ni boleh tahan sampai 2 bulan tau! [perhatian:agar2 ini tidak boleh dimakan ye... iyenye digunekan untuk mengculture atau dalam erti kate laennye... membiakkan bacteria~ heee~ ]
